CIK細(xì)胞最早是1991年由美國(guó)斯坦福大學(xué)生物學(xué)專(zhuān)家首次報(bào)道。他們發(fā)現(xiàn)在多種細(xì)胞因子(γ干擾素、CD3單抗、白介素-1和白介素-2)作用下，外周血淋巴細(xì)胞可以被定向誘導(dǎo)并大量增殖成為腫瘤殺傷細(xì)胞。 CIK細(xì)胞（cytokine-induced killer）是將人外周血單個(gè)核細(xì)胞在體外用多種細(xì)胞因子如抗CD3單克隆抗體、IL-2和IFN-γ等共同培養(yǎng)一段時(shí)間后獲得的一群異質(zhì)性細(xì)胞。CIK細(xì)胞兼具有T淋巴細(xì)胞的抗瘤活性和NK細(xì)胞的非MHC限制性殺瘤優(yōu)點(diǎn)，CIK細(xì)胞被認(rèn)為是新一代抗腫瘤過(guò)繼細(xì)胞免疫治療的優(yōu)選細(xì)胞。 同時(shí)CIK細(xì)胞是一種新型免疫活性細(xì)胞，它是自體外周血單個(gè)核細(xì)胞，經(jīng)多種細(xì)胞因子共同誘導(dǎo)培養(yǎng)產(chǎn)生的一類(lèi)CD3+和CD56+共同表達(dá)的高細(xì)胞毒細(xì)胞（殺傷細(xì)胞）。它是由美國(guó)斯坦福（Stanford）大學(xué)最先研制的，兼有T淋巴細(xì)胞強(qiáng)大的抗腫瘤活性和自然殺傷細(xì)胞（NK）的非主要組織相關(guān)性復(fù)合物（MHC）限制性殺瘤的特點(diǎn)，殺傷活性可達(dá)84.7%。CIK細(xì)胞制備流程

CIK細(xì)胞具有增殖快、殺瘤活性強(qiáng)、殺瘤譜廣的優(yōu)點(diǎn)，尤其是對(duì)化療耐藥的腫瘤細(xì)胞株亦有殺傷作用，而對(duì)正常造血集落的生長(zhǎng)沒(méi)有影響。通過(guò)取病人少量單個(gè)核細(xì)胞，在體外特定條件下制備出大量CIK細(xì)胞，回輸體內(nèi)后直接發(fā)揮有效清除腫瘤作用。

CIK細(xì)胞（Cytokine Induced Killer Cell，細(xì)胞因子誘導(dǎo)的殺傷細(xì)胞），是將人體外周血單個(gè)核細(xì)胞（PBMC）中的懸浮細(xì)胞在體外模擬人體內(nèi)環(huán)境用多種細(xì)胞因子共同培養(yǎng)增殖后獲得的一群異質(zhì)細(xì)胞，它具有顯著的識(shí)別和殺傷人體各種腫瘤細(xì)胞的活性。其治療可以在不損傷機(jī)體免疫系統(tǒng)結(jié)構(gòu)和功能的前提下，直接殺傷癌細(xì)胞，并且調(diào)節(jié)和增強(qiáng)機(jī)體的免疫功能，最大可能地恢復(fù)細(xì)胞正常的生長(zhǎng)調(diào)節(jié)，CIK細(xì)胞具有非MHC限制，具有廣譜殺癌細(xì)胞的特點(diǎn)，為徹底地進(jìn)行腫瘤治療提供了新的途徑。

DC細(xì)胞（Dendritic Cell，樹(shù)突狀細(xì)胞）是將人體外周血單個(gè)核細(xì)胞（PBMC）中的貼壁細(xì)胞在體外用多種細(xì)胞因子共同培養(yǎng)后獲得的抗原呈遞細(xì)胞，是正常人體內(nèi)存在的一種具有強(qiáng)大的抗原提呈功能的一類(lèi)特殊細(xì)胞，能夠直接攝取、加工和呈遞抗原，刺激體內(nèi)的初始型T細(xì)胞活化，從而誘導(dǎo)機(jī)體產(chǎn)生大量具有特異性細(xì)胞毒性功能的T淋巴細(xì)胞，對(duì)腫瘤細(xì)胞具有特異性殺傷作用。

CIK細(xì)胞、DC細(xì)胞可以單獨(dú)培養(yǎng)，也以共同培養(yǎng)成DC-CIK細(xì)胞，共同培養(yǎng)成的DC-CIK細(xì)胞可有效降低其中抑制性CD4+CD25+調(diào)節(jié)T 細(xì)胞比例，從而顯著增強(qiáng)CIK 細(xì)胞對(duì)多種腫瘤細(xì)胞的殺傷活性，起到1＋1＞2的效果。解放軍309醫(yī)院國(guó)際腫瘤治療中心同時(shí)擁有CIK細(xì)胞、DC細(xì)胞、DC-CIK細(xì)胞技術(shù)，已成功治療了近千例患者，取得了非常明顯的效果。
