Analyses of nano-UPLC eluteswere performed on a Q-Tof Premier mass spectrometer (Waters). Electrosprayionization was performed with an uncoated, pulled fused silica emitter (NewObjective) with a potential of 3.0 kV. The Q-Tof was operated in the V modewith a typical mass resolution of 10,000. Alternating scans were used to detectprecursor ions and then fragment ions. The masses of the precursors weredetected with a 0.5-s scan with a relatively low collision energy (CE) of 4 eV(MS scan), which was followed by a 0.5-s scan during which the CE was rampedfrom 15 to 40 eV (MSE scan). The instrument was calibrated with 13 fragmentions of the MS/MS spectrum of Glu-fibrinopeptide. To correct any shifts inmasses that occurred during instrument operation, a single-point calibrationwas performed against the lock mass compound, doubly-charged Glu-fibrinopeptide[mass/charge ratio (m/z) 785.8426], which was sampled every 30 s.
